چکیده

properties ofpolyene macrolides wehave established the structure-activity relationship among the derivatives of amphotericin B, the polyene antibiotic most commonly used in clinical practice11}. It has been shown in these studies that the improvement of selective toxicity is related to the absence of a free carboxyl group in the modified antibiotic molecule. Wehave also demonstrated that water solubility of amphotericin B derivatives occurs only with net charged molecules. In the present paper we report the novel group of polyene macrolide derivatives fulfiling the above requirements for improved selective toxicity and water solubility. These are polyene macrolide

Molecular modelling of membrane activity of amphotericin B, a polyene macrolide antifungal antibiotic.

Amphotericin B (AmB) is a well known polyene macrolide antibiotic used to treat systemic fungal infections. Despite its toxicity AmB is still regarded as a life-saving drug. The lack of adequate knowledge of the AmB mechanism of action is a serious obstacle to efficient development of new less toxic derivatives. Complementary to various experimental approaches, computational chemistry methods w...

A screening method for antifungal substances using Saccharomyces cerevisiae strains resistant to polyene macrolides.

Strains of Saccharomyces cerevisiae FL200 capable of growing on a solid medium containing a mixture of polyene macrolide antibiotics (nystatin, 40 micrograms/ml, amphotericin B, 40 micrograms/ml, pimaricin, 150 micrograms/ml and RP9971 antibiotic, 10 micrograms/ml) have been isolated after successive selection steps. The mutant strains, PR13 and PRC24, are 10 to 100 times more resistant than th...

Effect of fatty acids on action of polyene antibiotics.

Fatty acids cause a decrease in the absorption spectra of the antifungal polyene macrolide antibiotics nystatin, filipin, candicidin, and amphotericin B. For nystatin, filipin, and candicidin, this decrease in absorption could be correlated with the activity of the fatty acids in protecting the yeast Saccharomyces cerevisiae against the action of these antibiotics. With amphotericin B a correla...

YS-822A, a new polyene macrolide antibiotic. I. Production, isolation, characterization and biological properties.

A new polyene macrolide antibiotic, YS-822A was isolated from the culture filtrate of a mutant strain H-8 of Streptoverticillium eurocidicum var. asterocidicus S-822. Whereas the original S-822 strain produced not only YS-822 substances but also teleocidin as by-product which is well-known as a strong carcinogenic promoter, the mutagenized H-8 strain produced the antibiotic with only a trace am...

New N-alkyl derivatives of amphotericin B. Synthesis and biological properties.

Amphotericin B (AMB), Fig. 1 , the polyene macrolide antifungal antibiotic of high clinical importance, is the drug of choice for the treatment of deep-seated disseminated mycotic infections in humans1'2). However, the therapy is associated with toxic side effects, the most serious being the nephrotoxicity3). These undesirable effects are consequences of the poor selectivity of AMBinteraction w...
